چکیده

Introduction: Perchlorate (ClO4), a highly oxidizing compound, was identified on the martian surface by the Phoenix Lander at concentration levels up to 1.0% [1,2]. There is some debate as to whether an environment with perchlorate salts is too harsh for any organism to survive. Methanogens are able to survive in a numerous unwelcoming environments. Due to their ability to survive in extreme conditions, these anaerobic chemoutotrophs may have been the first autotroph to evolve on Earth, making them a good candidate for life on Mars [3]. In this project we studied the survivability and growth of methanogens in perchlorate solutions relevant to Mars [2]. Methods: Four methanogen species (Methanothermobacter wolfeii, Methanosarcina barkeri, Methanobacterium formicicum, and Methanococcus maripaludis) were used to test for possible methanogen growth in three perchlorate salt media (sodium, potassium and magnesium). The methanogen species were chosen due to their previous success of surviving in Mars-like conditions [3]. A specific medium was prepared for each species; CO2 was added as an energy source and H2 for an energy source. Perchlorate salt solution at different concentrations was added to the sterilized beakers. Tubes were inoculated with the appropriate organism and methane growth was measured by gas chromatography. Results and Discussion: Although methane concentrations varied with species and salt tested, all four species of methanogens produced detectable amounts of methane at all concentrations of each tested salt (Fig. 1). In all cases, there was little to no difference in methane concentrations at 0 and 0.1% perchlorate. In most cases, 1.0% perchlorate resulted in, initially, lesser amounts of methane. There are at least two possible explanations for this. The higher perchlorate concentrations may be inhibiting methane production by the methanogens. A second explanation would be that methanogenesis is not being inhibited, but the methane being produced is being oxidized by the perchlorate, resulting in less methane being measured in the headspace of the culture tubes. In some cases methane concentrations in the higherconcentration perchlorate tubes eventually reach the level of the control, supporting the second explanation. Implications for Mars: Although methane in the Martian CO2 atmosphere is globally very dilute, about Figure 1. Methane concentrations in Methanococcus maripaludis cultures exposed to three perchlorate salts. Diamonds represent the control, squares the 0.1% perchlorate, triangles the 0.5% perchlorate and the circles 1.0% perchlorate.

Effect of Liquid Media and In Vitro Pre-acclimatization Stage on Shoot Elongation and Acclimatization of Date Palm (Phoenix dactylifera L.) cv. Najda

The effect of stationary liquid media to improve shoots elongation in date palm cv. Najda and an in vitro pre-acclimatization stage to increase the survival ercentage of plantlets in the greenhouse was investigated. Two basal liquid media were tested: full strength and half strength Murashige and Skoog medium (MS). Media were devoid of plant growth regulators (PGRs), containing 0.5 mg/L 6-benzy...

Thermal and Evolved Gas Analysis of Magnesium Perchlorate: Implications for Perchlorates in Soils at the Mars Phoenix Landing Site

Introduction: Perchlorate salts were discovered in the soils around the Phoenix landing site on the northern plains of Mars [1]. Perchlorate was detected by an ion selective electrode that is part of the MECA Wet Chemistry Laboratory (WCL). The discovery of a mass 32 fragment (likely O2) by the Thermal and Evolved-Gas Analyzer (TEGA) provided additional confirmation of a strong oxidizer in the ...

Stability of perchlorate hydrates and their liquid solutions at the Phoenix landing site, Mars

[1] We studied the low-temperature properties of sodium and magnesium perchlorate solutions as potential liquid brines at the Phoenix landing site. We determined their theoretical eutectic values to be 236 ± 1 K for 52 wt% sodium perchlorate and 206 ± 1 K for 44.0 wt% magnesium perchlorate. Evaporation rates of solutions at various concentrations were measured under martian conditions, and rang...

Effects of explant type and growth regulators on callus induction in four ecotypes of Persian shallot

Allium hirtifolium commonly known as Persian shallot is an important wild medicinal plant distributed from North West to central and South West of Iran. To establish an efficient protocol for callus induction, the effects of explant type and growth regulators on callus induction in four ecotypes of Persian shallot were evaluated. Two explants types included basal plates and young leave...

The present study was carried out to evaluate the effects of light and plant growth regulators on callus induction and explant browning in date palm cultivar, Estameran in in vitro condition. The explants were pretreated by an antioxidant combination (100 mgl-1 citric acid and 150 mgl-1 ascorbic acid) for 3 days in 5 °C. Then, the  leaf explants  were cultured in Murashige and Skoog medium (MS)...
